I have isolated total RNA from blood samples of HBV by trizol method, converted it into cDNA and will now perform Real Time PCR for expression analysis of selected miRNAs by using the specific Micro RNA Primers. One of my seniors has performed similar work but she has used mirVana kit and isolated the Micro RNA content rather than total RNA content.
Which of these method is better and if the trizol method is not good, then will it give results or not for the particular MirRNAs?
I used total RNA isolated by trizol and miRNA enriched fractions for performing qRT-PCR of miRNA's. Although both of these methods were able to quantify miRNA's of my interest, Ct's were better when I used miRNA enriched fraction. Nevertheless, trizol gave the expected results.
