Note - there are variations and things that need to be adjusted as tissues can vary (whether you are using muscle tissue or skin tissue or fat tissue etc.)

Basic principle is to try to get rid of the unnecessary stuff (proteins and other things that can interfere with downstream reactions - clean them up and mostly using Polymerase Chain Reaction (to magnify the amount of DNA present before doing further tests).

Tip to optimize animal tissue dna extraction:

1) make sure to totally macerate the tissue

2) make sure you incubate long enough after adding in proteinase k, incubate overnight if required

3) after washing with wash buffer, try to heat the column at 50-60C to dry off all wash buffer residue

4) heat up elution buffer at 50C before use

5) after you pipette elution buffer into the column, incubate at room temperature for 10min before centrifuge, if you want your dna to be concentrated, you can reduce the elution buffer to 50ul instead of 100ul as suggested by kit protocol

or

if you want to combine dna from a few tubes, you can extract for example 3 tubes, you can run all 3 samples through one column so that dna from all 3 tubes will be concentrated in one column, then proceed with extraction protocol.

Hi Muhammad, you will not find any problem. If the DNA is pure (even though little amount) it will be dissolved in water when you add it to the tube. Nevertheless, I think if you got a very low yield you should start from the beginning taking more amount of sample and do as Vikki recommended.

Hi Muhammad, while you probably should restart your DNA extraction, if it's impossible for some reason. There is no major issue with drying up the DNA and resuspending in a lower volume. We routinely dried up our oligos or plasmids with a speedvac to keep them dried at -80C for extended conservation (5+ years) and they would resuspend very well. Typically, we would add up to 50% of final volume, vortex heavily for 30 secs, let rest for 30 secs, vortex again for 30 secs. Wait 15 mins and then complete to final volume and vortex again for 30 secs. Everything should be back in solution.
