Dear researchers,
I am facing problem in maturating my OPCs into mature oligodendrocytes.. I have been using T3 and T4, also adding NAC and CNTF in the maturation media, even after 2 weeks I dont see the protrusions I suppose to see. I am using pig cells.. In 6 well plates which made up of plastic, there are more protrusions than that of glass chambered slide I have been using so far. Does anyone have any input please?

Hi Goutam,
Cells in vitro can be a little difficult to characterise by morphology - and pig OPCs are not very commonly cultured. Have you tried staining them for MBP to assess their maturation?

Many thanks for your quick response. I am using GalC and CNPase antibodies and they look fine after 2 weeks of induction. But I dont see nice morphology with amazing oligodendrocitic protrusions.. Some of the cells do have those morphology.
