Dear Colleagues,
I have started growing SLICE Culture in our laboratory.
I dont have any reporter gene cloned, I would like to ask, how do I make sure that the culture is growing and the cells in the slice are not dead?

Hi,
I did a lot of brain organotypic slices and usually we performed a follow up among time of different markers such as DAPI, for total cell number, or NeuN, Tuj1 or TH, to check neuron survival. Then we performed a quantification of those marker among time to have an idea of the survival of the slice. Of course you should adapt the choice of marker to you project. In a general manner, brain organotypic slice don't survive more than a few weeks.

Hello, this paper has some excellent measures of organotypic culture vitality. Including metabolism rate assay, LDH assay and immunohistochemical markers for neurons and glia. Best of luck with your cultures.
