Hi, I also use Trizol for cell homogenization but i think the lymphocytes count you are using is quite low. If your white cell count is higher, say 8x10^6 cells or more, the RNA yield will be higher.

I am using Qiagen RNA extraction kit and so far it has been giving us good RNA yield and quality.

• I too have used both Trizol and RNEasy kits for blood from qiagen to isolate blood from PBMC
• I have used both with success but like Zx your starting cell number is low so you wont return much RNA
• You can lose RNA during Trizol chlorform extraction so I would and do opt for a blood specific RNeasy kit when starting with a small amount of material like this

The cell number should be sufficient. We use Trizol and it works well. Your RNA seems to be good quality. It depends on your downstream application whether the yield is sufficient. You have enough to do several RT reactions.

Trizol typically works great, for all sample types including cells. for higher purity Trizol plus (with downstream purification column) can be used. Alternative option for cells is to use Cells-to-Ct kits - if your downstream analysis is qRT-PCR. in this case, there is no RNA purification: cells are lysed, and analysis is performed straight on crude lysate - which is fast, and there is absolutely no RNA loss "on the way"
