I would like to know if we can measure E3 ligase FBw7 activity or protein expression in breast cancer cells following drug treatment.
Can you just do an in vitro ubiquitination assay and then separate your protein by SDS gel and then do a western blot to measure the intensity of the smearing using anti-Ub antibody?
Check one of our publication which is describing that but with SKP2
http://www.ncbi.nlm.nih.gov/pubmed/12769844
and also Markus Welcker 2004 he has been working with FBW7
I would be grateful to receive recipe for extracting total protein from breast cancer cell lines to see expression of phosphorylated proteins.

I was reading a paper on Leukemia Stem cell and could not understand why mice were exposed to radiation before inoculation of CB lin- cells and surrogate neoplastic stem cells.
