I want to isolate single-stranded DNA from a mixture with double-stranded ones in a reassociation experiment. Any help will be greatly appreciated. Thank you!
You may try a classical and reliable Exonuclease III (ExoIII) from E.coli commecially available from most biotech companies. Exo III is an enzyme that belongs to the exonuclease family. ExoIII catalyzes the stepwise removal of mononucleotides from 3´-hydroxyl termini of duplex DNA. The preferred substrates are blunt or recessed 3´-terminy of any DNA. Importantly, the enzyme is not active on single-stranded DNA, and thus 3´-protruding termini are resistant to cleavage. Best wishes ALes.
