Anyone working with the amplification of ITS1-5.8S-ITS2, how may bands do you usually get using primers that span from the 3'-end of 18S and the 5'-end of 26S? I got 2 bands from my sample and three bands of my other sample, all from the same species but different lines/varieties.

I think there could be variation in the ITS1 or ITS2 lengths giving rise to the variation in amplicon size. I just sent my sample for sequencing.

Can you share experiences about the usual number of bands you get?
