I inoculated E. coli O104 in orange juice in a flask and kept it in the fridge in front of which there is light that lab personnel commonly use while working in lab at least 12 hours. Now I am worried whether this light exposure affected my inoculated bacterial growth or not? if affect what I can do in this case, anybody knows, could you please suggest me?
Azimun, I really do not know. But have you looked at it under a microscope? And what kind of light was it? To my knowledge only UV ray lights would kill E. coli, and that would require direct exposure, not through a bottle or other material.
@Chartrand, Thank you very for your answer, I would like to say that I already mentioned in my question explanation that I inoculated bacteria in orange juice in a Erlenmeyer flask which is getting exposure daily to visual light since I kept the flask in fridge which is infront of visual light.
The type of light could still be important - is it mostly blue light? Is it red? Indoor lighting really shouldn't have an impact on E. coli growth but it's easy enough to test. If the fridge gets opened a lot then the change in temperature or humidity is likely to have more of an impact than light exposure.
All good points, Harry. I would think verification for the answer to her question would still require microscopic observation, also.
@Thanks all, I would like to inform you that the fridge I used , the door of which is made of glass and door always remain closed and I am talking about visual ( white light) light but not other color or UV light.
I cannot imagine that white light through a container barrier would have any effect on E. coli. UV light would, but then, it would be safe for human eyes without protection.
There will not be any effect of visible light on the growth of E coli. However, you should look the effect on orange juice and some anti-oxidant produced from the juice that can cause any effect. regarding the UV, there is is no question of UV here, even then, since the container is glass, UV cannot penetrate them, so there will not be any effect. Moreover you will get very less growth due to the low temperature in the refrigerator. In general I support the point of Dr. Chanrtrand that you have to do microscopic or may TVC to check the effect.
