I'm having some trouble with IC50 dose calculations. I try to do differently in Prism, Excel, etc. and the results are not matching sometimes.
And sometimes my dose-time plots do not show a steadily decreasing viability, causing higher and lower than 50 values to be more than one. If I need to speak hypothetically it can be like;
1 ug/ml: 55%
2 ug / ml: 47%
3 ug / ml: 53%
In such cases, I am thinking of remove some doses, but I do not know how accurate this is and which one should I eliminate. Why are my graphs coming out this way and are there any tricks for the IC50 calculation? I will be happy if you can help me.

1. As you are using prism for the calculation of IC50.
2. Prism also provides helping literature about the analysis (how to do), you can go through that also.

Thank you for your answer, but as I said, the main problem is that my responses to dose are not in a constant decrease. I don't know how to act in this situation. Even though I tried several times, my results came out like this.

You can try with ten different concentrations and see the trend if still it will come then choose five or six out of ten by taking the lowest and highest same concentration.
I hope it will work for you.
e.g.
If you are taking initially 1,2,3,4,5,6,7,8,9,10 ug/mL.
Then if needed to adjust then you can go with 1, 2, 4, 6, 8, 10 ug/mL.
Moreover, if you are making the drug stock solution in any vehicle alone then you can try making it in culture medium (If applicable in your study).
One more thing you can try is to go with higher concentration to check the handling error.
e.g. 10,20,40,60,80,100 ug/mL and still more if needed.

Actually, I've worked with a wide range of doses, including very high and very low concentrations, but I don't know which doses to eliminate or if it's right when the result is like this. After all, I'm working iteratively and this is the data it gives me. Also, I had prepared my drug from the main stock in the medium. When I repeated later, I saw similar results. Maybe it has to do with the drug's mechanism of action.
