I need to study point mutations in a gene in order to understand its function in response to certain stimuli. So, i want to create a knock-in of the mutated gene by CRISPR/Cas9 via Homology-directed repair method.
Does the THP-1 have this gene? If so, why don't you just knock-out the gene directly using CRISPR/Cas9 to study this gene's function?
I'm trying to understand point mutations of the gene with respect to the function of the gene. I already know what happens when we knock it out.
