I am working with a synthetic compound that can induce DNA fragmentation in MCF-7 and MDA-MB-231 cells. When I co-incubate with z.vad.fmk, fragmentation is only partially inhibited in MCF-7 cells and fully inhibited in MDA-MB-231 cells which suggests that activated caspases take part in this process. However I can only detect PARP cleavage (89 kDa) only in MCF-7 cells but not in MDA-MB-231 cells. Is this possible?
