Are there any conditions where it would be possible to lyse gram-negative bacteria using the Promega PLB?
I would need to lyse the cells to test for crude enzyme activity but I don’t have a protease inhibitor cocktail. I’ve read that PLB has protease inhibitors but they’re optimized for lysing mammalian cells. Would it be possible to lyse bacterials cells using this? Has anyone tried this?

I don't know what PLB is composed of but probably just a detergent. It might work for gram negative bacteria especially if you included some freeze thaw cycles. However you can just do the same with any non-ionic detergent, like Triton X100 at 0.1% (I would not use SDS). For most well folded proteins that are enzymatically active you don't really need to worry about protease inhibitors, you should still have activity although there may be some proteins that are rapidly degraded.
