I am running MTT cytotoxicity assays on marine mammal skin fibroblasts. Although my results have been good for many toxicants, PCB126 results have been poor. I run my assay in triplicate and in each assay at least one or two columns have CV% values above 15.00, which is unacceptable. I am not noticing differing levels of cell die off between wells under the microscope so this is perplexing. Variability of dosing between wells in each column is highly unlikely as toxicants are vortexed with media before being used. I am using a cell line from a single animal for this particular set of experiments. Has anyone else had higher than acceptable CV% values with a certain toxicant while other assays turn out acceptable? Any advice would be greatly appreciated.
