I have to culture splenocytes and retrogenic TB10.4 CD8 T cell clone separately. I was wondering if I should add sodium pyruvate to the RPMI 1640 medium. I usually add 1 mM sodium pyruvate to the 500 ml RPMI bottle for culturing splenocytes but am not sure if I should keep the same medium formulation for culturing other cell types. Please suggest.
Pyruvate is a metabolite of glucose, which I agree that is the main source of energy for the cells. However, sodium pyruvate can be added as an extra source of carbon, without causing any harm to the cells, I agree. Regarding the necessity of adding it to your cell culture medium, I would say that it depends on your question of interest. I would suggest to add sodium pyruvate in case you would like to help the cells to defend themselves against oxidative intermediates. (Therefore, please be careful with performing any ROS assay while having the cells in a medium containing sodium pyruvate.)
sodium pyruvate solution added to cell culture as an additional source of energy. it may help low density cell grow & produces energy in the form of ATP. it will not effects any cells. you can use 1mM sodium pyruvate any cell type.
We have been culturing Splenocytes or Peripheral blood lymphocytes in RPMI-1640 (10%FBS and Glutamine) and they grow well. We do not supplement this media with sodium pyruvate as an additional source of carbon. RPMI already contain glucose which is major source of carbon for cells. Cells also have an inherent property to produce sodium pyruvate- glycolysis intermediate. There is no harm in adding additional souce of carbon.Also if cells have been grown in medium that is supplemented with sodium pyruvate then it is recommend continuing its use as cell growth may lag without it
RPMI contains 11.1mM concentration of glucose. I always use 2g of sodium carbonate which I think is ok as prescribed, with 2-3g Herpes.
