What is the ideal solvent and concentration in which rosuvastatin-calcium can be dissolved, for cell-culture applications?
Is it soluble in water or do we need ethanol or DMSO solutions? If so what should be the concentration of Ethanol/DMSO for dissolving rosuvastatin and still be able to be applied on mammalian cells?
Soluble in water (< 1 mg/ml at 25 °C),
chloroform, DMSO (100 mg/ml at 25 °C),
methanol, and ethanol (< 1 mg/ml at 25 °C).
You can use ethanol as 5% or you could use phosphate buffer pH=6,8 to desolve the statins.
For my previous research, I used DMSO to dissolve rosuvastatin as it wasn't soluble enough in water for me to achieve the concentration I needed.
I would recommend not going above 0.1% DMSO in cell culture medium, as at higher concentrations DMSO can be toxic to cells. I did this by initially making up a rosuvastatin stock solution of 100 mM in DMSO, and then diluting this by 1:1000 in the cell culture medium, i.e. the highest final concentration I was able to achieve in my cell culture experiments was 0.1 mM. I hope that makes sense, but do ask for clarification if you need it.
