I have DNA template which have 2 restriction enzyme sites but I wanna change these restriction sites to another restriction enzymes, can I design new primers with the new restriction sites I want and use that old DNA template?
You can clone the DNA template a vector with present restriction enzymes and cut again other available enzymes you want to use.
you can design pcr primers with the new cut sites ( add 3 extra bases to make cutting easier) to amplify your target. If you need to get rid of the original cut sites just incorporate a suitable mismatch if the primers cover the original cut sites and adjust the annealing temperature appropriately
