Hi, I am studying the interaction between nanoparticles and cells that are grown on hydrogels (VitroGel). I want to perform a TEM characterization, however I have issues in finding the right protocol for epoxy resin infiltration into the matrix. (please, note: I am willing to do a RT sample preparation) I tried to infiltrate epoxy resin using either ethanol or propylene oxide, but it didn't work. I am thinking to try with acetone too.
Do you have any suggestions/protocol you'd like to share?

Dear Paola, your question is very challenging. Probably the infiltration of the hydrogel matrix (that is hydrophilic) by some other solvent than aqueous solutions will not occurs. You can try a gradient of solvents (change the aqueous solution using different mixtures of water/solvent) or try to fix the cells on the hydrogel matrix using glutaraldehyde and then dry the sample (if possible of course). Cryo-TEM is another option (again.. I don´t the robustness of your system for this). AFM analysis is not helpful?
