My final research is about making chitosan/pva hydrogel for wound healing using gelatin as crosslinker. Last month, i made the thin film hydrogel and it turn well. It was okay. Last week, i make it again with same ingredients and protocol, but why my hydrogel dissolve in PBS and form a honey-like consistency? Does anyone know the reason? I would love to hear disscussion from you guys. Thank you!
The coacervate complex has become soluble - before doing anything else check the ionic balance and pH. Coacervates can be solubilised unexpectedly if the ionic strength or pH are borderline.
One reason could be the disassembly of the gelator molecule in the pH. It generally happens if you are using a gelator molecule that is prone to protonation.
The dissolution of your chitosan/PVA hydrogel in PBS could be due to insufficient crosslinking, variations in composition, unfavorable environmental conditions, or inconsistent preparation methods. To address this issue, ensure proper crosslinking, maintain standardized conditions, and conduct stability tests to evaluate the hydrogel's performance before use.
The dissolution of your chitosan/PVA hydrogel in PBS, forming a honey-like consistency, could be due to improper crosslinking. If the gelatin did not effectively crosslink the chitosan and PVA, the hydrogel would lack the necessary structural integrity to remain stable in PBS. As you mention that the your hydrogel was good last time, I think in the second attempt there is lack of the crosslinking in the gelatin.
