I am currently using Qiagen micro/mini RNeasy extraction kits to extract RNA from primary mouse DCs following 6 hour incubation in culture media in order to perform RNA sequencing. However, compared to when I do it with BMDCs, I get really low concentrations and the integrity is pretty poor. As far as I know the cells aren't dying during the incubation, does anyone know of a method or a different kit to use which would yield better results?

I am not sure if it would help as I used it with lichen material, but I found that Trizol reagent from Life Technologies worked fairly well, with pretty good yields and medium to high quality. It worked perfectly for my reverse-transcriptase PCR, so overall I was very pleased with the product (Trizol).
trizol works really well for isolating RNA from most kinds of biomaterials. i have tried that for plants, filamentous fungi as well as from animal cells. i have isolated RNA using Trizol with RIN value upto 7.5 from filamentous fungi and upto 7 from mammalian cells.
