There are a lot of nonspecific fluorogenic protease substrates. We conjugated FITC to BSA and really saturated it which quenched the fluorescence. When hydrolyzed, there was a dramatic increase in signal.
Just looking around, ANASPEC has a substrate used for trypsin, cathepsin, etc. AS-60323. If you think it doesn't work with papain, look around. There are lots of similar offerings.
