Hello Everyone, I'm going to isolate exosomes using ExoQuick from 10 ml of culture media, by using 1/5 amount of the kit. After isolation how much PBS should I dilute to preserve the exosomes.
I used 500 uL of Crevicular Gingival Fluid (FGC) with 120 uL of precipitation buffer of exoquick and finally to concentrate the sample (because the kit is not made for the to this type of sample) I dilute in 50 uL of PBS1X (without tween, because of the degradation of exosomes).
Making the calculation for your volumen of sample, you should resuspending in not less than 500 uL.
Thanks a lot Ana.....But the last sentence is not clear to me ...you said you have resuspended in 50 ul and then you mentioned not to go for less than 500 ul..
What I meant is that the initial volume of my sample were 500 uL unstead yours are 10 mL. I only did the calculation using my volumes using standardized protocol for FGC.
FGC: 500ul----- 50 uL PBS1X
Culture media: 10.000ul----- x =
x = 1000 ul
x=500ul (if you want to concentrated even more your sample)
I did kinetics volume to resuspend according to the amount of protein sample obtained ,because as I commented this kit is not made to the sensitivity of such a specific fluid in which I worked
