I am planning to incorporate 1 mM L-homopropargylglycine into HeLa cells in methionine-free DMEM media. I know that the incorporation of this alkyne-containing amino acid into nascent proteins occurs within 1-4 hours of treatment. However, how long does it take for the cells to then clear these alkyne-containing proteins from the cell? The longer they can be retained the better, even up to several days if possible. Any recommendations or information is welcome.
