I am planning to incorporate 1 mM L-homopropargylglycine into HeLa cells in methionine-free DMEM media. I know that the incorporation of this alkyne-containing amino acid into nascent proteins occurs within 1-4 hours of treatment. However, how long does it take for the cells to then clear these alkyne-containing proteins from the cell? The longer they can be retained the better, even up to several days if possible. Any recommendations or information is welcome.

Depends on the cell type and the protein your analog has been inserted into. Some proteins have a fast turn over while others can take years.
