Hi,

I started culturing some ipsc-RPE cells in Vitronectin XF coated 24-well plates. I've noticed that cells start detaching and and dying in areas of the well 2 days after plating.

I only just realized realized that I've been using the a 1:25 dilution of Vitronectin, which is the recommended dilution on the Vitronectin XF stem cell protocol, however, the ipsc-RPE plating protocol calls for a 1:100 dilution.

Do you think this might be the reason why the cells are the dying, or have people used a 1:25 dilution on ipsc-RPE cells successfully before?

Just trying to figure out if perhaps I am missing something else. Thanks!
