We are conducting experiments to detect inhibition of migration in U2OS and Saos2. We detect an increase in e-cadherin mRNA but we are not able to detect the corresponding e-cadherin protein on Western-blots. We have tried with other cell lines and the antibodies work fine. Please, does anyone has similar problems?

Hi Mozart, to make a case for E-cadherin in migration (or lack thereof) you will probably have a stronger case if you can do fluorescent microscopy to show that E-cadherin is specifically localized in cell-cell contacts and not as a non-localized punctate pattern.
