I know how to separate pbmc's from blood, but I am just wondering what the most efficient way of doing so with 50ml's is?
I have a rotor for my centrifuge that fits 15ml and 50ml tubes. So if I'm separating pbmc's from 50ml's of blood, I would place how much blood into the 50ml tubes along with how much ficoll/density gradient? I am just trying not to waste tubes and time centrifuging small amounts at a time.
Also, if you could be so kind as to give an estimation (either on volume in ml's or cell count) on how much pbmc's 50ml's of blood will yield.

I used to get 60ml blood, dilute 1:1 with PBS and load 30ml of diluted blood onto 15ml of ficoll in 4 x 50ml falcon tubes. I would get around 50-60 million cells
