With powdered plant, you can use Soxhlet apparatus (heat treatment)
Or you can use the plant extraction at room temperature in a recipient, but it takes more time (you have to change the solvent if it will be concentrated)
powdered plant, may be extracted by using soxlet extraction method solvent extraction as well as by using different surfactant .if u required i wll send you my paper for detail methodology
Just put your plant material in petroleoum ether and let it stand for sometime. Shake or keep it in water bath at upto 40 deg. and after that filter and evaporate solvent. Residue you get is pet. ether extract of your plant. or if you have a soxlet apparatus then thats better.
Kindly, I am trying to test the MIC from the leaves using microdilution method. However, the extract is green due to the chlorophyll. I need to remove it. I tried active charcol but the green color didnt been removed. Any other way ?
Also I will use microplate spectrometer , so I am wondering if it is a correct metgod to read the MIC of the plant extract ??
We use active charcoal to remove the chlorophyll usually it works .Use appropriate amount of active charcoal for it.Or you may go for Column Chromatography .
I dont have Column Chromatography. How much should I add to the extract ?
What I did was after the extraction , I added around 50g of activated charcol to 300 ml extract and then filterate it. But the extract was still green !
Is this a correct procedure ? Should I wait for specific amount of time ?
