I want to analyze the iron nanoparticles I synthesized using DLS and zeta potential methods. I did that analyze before, but this time I want to do DNA-binded nanoparticles. I read a few papers (in below) but I don't know how exactly I'll do that DNA-nanoparticle complex. Who has an idea on this subject? Thanks a lot!
Hello Sedef Ozel , if I understand your question correctly, you have already been able to measure DLS and zeta from iron nanoparticles. Now you created a new set of samples which are a complex of DNA+nanoparticle and you want to measure that sample. Do you already have any data you can share? Have you tried the same light scattering method as with your iron nanoparticles?

That's right! Actually the part of I don't understand is how can I create DNA-nanoparticles complex. I mean, what DNA should I use? When can I add DNA to the nanoparticle? Is it just before the zeta potential measurement, or do I need to make a preparation earlier? Thank you!
depends on what exactly you want from the zeta potential analysis.
if the goal is to find out how DND is adsorbed onto the surface of the particles, then use the time protocol on the zetasizer. set the measurement of the zeta potential every 5 minutes (for example) and the duration of measurements is no more than 2 minutes (for example). and analyze as much as you like in time. at least 1 hour, at least 2 hours. do not use very high concentrations of DNA to avoid sedimentation. or vary the acceptable DNA concentration in advance.
if the goal is to find out how DNA is adsorbed onto different (for example) particles for a certain time, then add DNA to the particles, wait 5 (for example) minutes, then rinse if you want, then measure. for another type of particles, the same protocol.
