Is there any method for the long term storage of Acidithiobacillus species as we need to subculture it every 2 weeks?
At. ferrooxidans can grow suboptimally at pH 3 or 3.5. At that pH agar can solidify but you have to autoclave it separated from the rest of the medium and mix all together when they are about 50-55 °C right before pouring plates

We can prepare glycerol stock for Acidithipobacillus ferrooxidans according to some papers i have studied it is not possible to store it for more than 2-3 month and this culture require subculturing on every 2 weeks. Solid media preparation is also difficult as it require very low pH for growth and at that low pH solidification of agar is not proper. As per your suggestion I will definitely try for semisolid media and glycerol stock preparation. Sir it would be very nice of you if you can suggest me some solidifying agents that can withstand such a low pH and cheaply and easily available.
