We can prepare glycerol stock for Acidithipobacillus ferrooxidans according to some papers i have studied it is not possible to store it for more than 2-3 month and this culture require subculturing on every 2 weeks. Solid media preparation is also difficult as it require very low pH for growth and at that low pH solidification of agar is not proper. As per your suggestion I will definitely try for semisolid media and glycerol stock preparation. Sir it would be very nice of you if you can suggest me some solidifying agents that can withstand such a low pH and cheaply and easily available.
