I'M working on the soil Enzymes. I need to to check the total reductase enzyme production and its activity produced by certain enzymes especially when the inoculum is converted to liquid media. Can anyone suggest to me a particular method or protocol to analyze the reductase enzyme production by the microbial community in the media?
You need to be more specific. Which particular reductase specificity are you interested in? Nitrate reductase, aldehyde reductase, or any of about 500 others?
If you have a specific assay in mind, then the normal approach would be to make a soil suspension in an appropriate buffer (possibly including a low concentration of detergent and/or a short sonication to dissociate cells from soil particles), centrifuge at moderate speed (to remove soil particles but not cells) and then use a small aliquot in your assay (suitably replicated and with controls, including a heat-inactivated sample).
