For attachment of plga to amine modified aptamer, we need carboxylic acid terminated PLGA, but we have ester terminated plga.
1. But now, i don't know can i use the commercial methods of ester conversion to carboxylic acid for this purpose ? like H2o/THF mixture with LiOH or KOH-methanol or other suggested methods.
2. since the other terminate of our plga is alcohol , can i use the commercial methods of alcohol conversion to carboxylic acid like
Potassium dichromate ?
3. could used these methods for this purpose without change of plga nature for last reactions?
4. which one is better changed ? protein encapsulated plga particle or plga before preparation
The easiest thing is to buy acid terminated PLGA. The ester conversion to carboxylic is probably harsh for the polymer. For the hydroxyl terminated plga you could use N,N'-carbonyldiimidazole.
I would recommend to purchase PLGA-acidic. you can also request a gift sample to the provider in your region.
