I want to block a receptor on the cell surface and then treat the RAW cells and I wonder if I can used a specific antibody on the cell culture. Did anyone ever do that?
I have done this previously with the EGF receptor using Cetuximab. You can do this if you can find a receptor inhibitor which functions in this way. My suggestion would be to look out for any drugs used to inhibit your receptor which ends with -imab. Any drugs with this at the end will be antibodies and if they work on a receptor it is likely they act competitively to block ligand binding.
I have done this with a specific antibody for EGF receptor and for P2X7 receptor. In an other experiments I have used a chemical inhibitor for both EGF and P2X7. You can also transfect cells with a siRNA to knockdown the receptor in your RAW cells. I did this using a siRNA to block P2X7 receptor in HaCaT cells and then after the knockdown I treat the cells with the drug I wanted to test.
