I am working on metal oxides. I need to make some conjugates of drugs and NP. But I am unsure of what concentrations to make. I have read some articles, but they haven't mentioned how many concentrations of NP and drug to be added to make those conjugates.
Secondly, I am doing just in-vitro activity using the cell lines. So is it necessary to coat those NPs? I need help.
Dear Sarah,
First of all, I reccomend to do an in vitro experiment on your cells by adding different concentrations of your drug to see the cytotoxic effect on your cell population. Depending on the drug, you could start by adding 0.1 to 5micromolar concentration of your drug (do at least 8 different concentration in this range) and measure the cell death after 48 or 72h. This experiment will allow you to determine the ED50 of your drug for your cell line (the concentration of drug you have to use to cause 50% of cell death). When you know the ED50 concentration, you can then add your drug to nanoparticles and quantify the drug loading. Then you can repeat the same experiment I explained before with your nanoparticle-drug conjugate and compare the effect to the free drug (using the same amount of drug conjugated to your nanoparticle and free drug in different wells to see if you can improve the effecacy of your drug by adding it to the nanoparticle or not).
