Macrophages are notoriously difficult to transfect. I'd like to be able to screen a handful of constructs for their effect on cytokine production in macrophages before selecting a couple for more extensive and publication quality experiments using vpx lentivirus infection. Any and all tips appreciated. Thanks!

for transfection macrophages I use a new transfection reagent called Viromer.
Its a synthetic polymer zero in charge. Due to an active endosome escape mechanism I get higher transfection efficiency compared to standard reagents.

Which conditions of the protocol did you use? For plasmid transfection please find attached a protocol from one of our customers being very happy with the efficiency.
