Cytotoxicity from your transfection agent can be an issue. I don't have experience with Gastric cell lines, but have you considered using a different transfection agent (e.g X-tremeGENE 9)?

Thank you all, yes i reduced the quantity of Plasmid as well as Exposure of PEI mixture from 5 hours to three hours..result has been improved but need more optimization.
