May someone suggest a comprehensive review on how the expression of two forms of a single miRNA 5p and 3p (formerly known as miR and miR*) is regulated in a single tissue? I am willing to know more about the general principle involved in this (if there is any), rather than the individual regulation of expression of any particular miRNA.
Hi I am not sure what review to direct you towards, however I have read intensively on this myself. Firstly, different microRNA have different levels of stability and this depends on the 5' end of microRNA and its adherence to the target in RISC complex. So how strongly a particular microRNA 3p/5p binds in risc is a major judging factor. Also some microRNA such as mir-107 are actively degraded when not required.

Thanks for the reply. I was wondering what mechanism decides which form (that is either 3p or 5p) is to enter RISC complex and which one to degrade? I understand that it is not absolutely determined which form (5p/3p) will enter RISC complex in which case. I also understand that it varies species-wise and even tissue-wise. What I am trying to understand is the decisive mechanism behind the issue i.e. which form will bind to RISC and which one will degrade. Is there any particular pathway/protein complex involved in this?
it depends on how strong the base pairing is. The stronger one stays and the weaker one is degraded.
