In my research, over 70 bacteria strain was isolated. Now, I need to use PCR method to find 2 group of Bacillus spp. and Non-Bacillus.
Could you suggest me which specific primer to identify Bacillus spp., please?

You can use the specific primer (universal 16S primer). Also, this primer is used to differentiate between species of Bacillus.

To select species specific primers find attached an SOP I devised for students some time back which describes in detail
1. Using Ensembl/Genbank to identify target region of interest
2. Designing primers to resulting target
3. Do's and dont's pertaining to optimal primer design
I realise this is general but nonetheless I hope you find it useful
Samy M Sayed You are wrong. 16S can not resolve the species in bacillus species (with confidence). Moreover, you are suggesting universal 16S primer, absolutely wrong.
