Dear researchers. I tried using the IHC PROFILER in image j to quantify nuclear DAB staining. I followed the instructions in the original article by "Varghese F, Bukhari AB, Malhotra R, De A (2014) IHC Profiler: An Open Source Plugin for the Quantitative Evaluation and Automated Scoring of Immunohistochemistry Images of Human Tissue Samples. PLoS ONE 9(5): e96801. doi:10.1371/journal.pone.0096801".
I find it difficult to identify the nuclear positivity when working with "threshold" tool. very grainy areas appear- not representing the nuclei as nuclei. instead too many dots are seen in each nucleus.
I use images clicked in 40x magnification from human oral squamous cell carcinoma tissues. Esteemed researchers who have used this method of quantification can shed your experiences. kindly be moderately descriptive in your response. Thanking you in advance.
