Dear scientist,
Am having a small molecule ( selected by in silicomethod) to target a gene or protein (x) which involves in cell division of bacteria.
1) I want to prove that the small molecule binds to that particular Protein(x).
2) As well as the small molecule have the effect on cell death of a bacteria.
i have decided to do MIC and Kirby bauer method for the 2 condition.
But to prove the 1st concept, i feel like cloning the gene into a vector and express it high and see the effect of small molecule on the cloned bacteria and purify the protein from the cloned bacteria before and after treatment with small molecule.
Am i right with my protocol ? is there anything else needed to prove the effect of small molecule on the bacterial cell division defect?
kindly suggest me a good protocol as well as some article references.
