Absolutely no. Self-renewal is one of two fundamental properties in stem cells. To maintain that properties, they should continuously be fed with serum, specially many unknown factors of FCS to achieve efficient proliferation. There are few reports that claim chemically-defined or xeno-free media with defined GFs mix or low molecular mass of peptides mix. These studies are not widely validated by nobody yet, just like many degradable plastic bags' claim. Practicability, efficiency, speed, and reliability etc would not allow FCS replacement yet. With stem cells i do not recommend omitting FBS and growth supplements from media where media are optimized with these components in order to support stem cell growth , Self-renewal and survive .
yes, there are plenty of alternative to cultivate stem cell without serum.
for instance, for mouse ES, you can use KOSR (knock out serum remplacement from Invitrogen) coupled with KO DMEM, or even the 2i medium.
for human IPSC or ES, you have as well a lot of alternative media without FCS that are used in combination with various matrix. Indeed very few lab still cultivate hiPSC in FSC. too much trouble to get a good batch.
it depends what you constitute as fbs/serum free. Embryonic stem cells tend to use conditioned media whereby knockout serum replacement (20%) along with knockout DMEM, 1% L-glutamine, 1% NEAA and 4ng/ml bFGF which is conditioned on MEF for 24hrs and then filtered and again supplemented with another 4ng/mL bFGF.
when we isolated hMSC we used DMEM high glucose, 5% FBS (batch tested), 1% L-glutamine,1% NEAA And depending on oxygen environment would dictate the population doubling capacity. We would not supplement the hMSC with bFGF.

I am rather shocked that anyone would claim that stem cells cannot be grown without FBS. Both human ES and iPSc cultures are routinely done without FBS. The two most commonly used brands of media used for ES and iPSc culture are mTeSR and E8 media. Both of them are serum free.
At least some types of stem cells can be, and are, cultured without FBS.
Our hMSCS are growing better and faster with commercial xeno free medium PRIME XV from Irvine. Especially for MSCs there are PLENTY of commercial serum free options out there!
