We are standardizing a method to Hybridize DNA from Colonies onto Nylon membranes. We already have two protocols to try, but what I would like to see at the end of the "try outs" is if the DNA: 1) Bound to the nylon membrane, 2) Good ammounts of DNA were collected.
I was thinking to stain the DNA with a general DNA stain like DAPI, Propidium Iodine of Sybr-Green and look at the membranes on our scanner for fluorescence. Does anybody has experience with this?
Hi Luisa. If your nylon is infrared compatible, then Syto dyes can be used to scan the blots directly on a Licor Odyssey (700nm channel).

We tried a couple of months ago with PeqGreen and it works. sadly we had VERY little DNA hybridized. I think it should work as well with the syto dyes.
