I need to do anticancer activity and some biochemical parameters of plant extract in mice, my extracts dissolves well in pure DMSO. Is it safe to use pure DMSO as a vehicle if not, what percentage of dilution is normally recommended?
No. DMSO should only be used less than 2% for application. Reason: DMSO changes membrane fluidity and transmission, both of which can drastically Change the results and may be toxic to some extend.

I've seen DMSO being used as transdermal vehicule, but it's really not the best vehicule for PK studies. DMSO is as CYP inhibitor so it can change your clearance quite a bit. If you use DMSO, the rat/mice tends to "stink" as it swells DMS from reduction of DMSO (DMS, DiMethylSulfide, has a quite strong stench...).
thank you all for your valuable answers. my extracts dissolves well in DMSO but now i think i should search for some other vehicle. is it ok to use solvents like carboxymethyl cellulose in 0.9% saline even if the extracts don't dissolve completely for intraperitoneal injection?
transdermal or i.p.: go for it, but follow the restricitions people above have mentioned.
intravenous: forget the DMSO, try ethanol or albumine-solutions (in water) to get it soluble. It is to toxic, side-effects may hurt your experiment(s).
