Yes, it is possible. Of course your column needs to be able to withstand whatever back pressures are developed. Columns packed in stainless steel are available commercially from several manufacturers. Check their web sites to find a column suitable for your size range.
You can use SEC or GPC columns in a HPLC system. However, you can`t call this method as high performance-SEC, because the performance of the separation depends on the column, not the LC system employed.
A comment to Ms. Bano´s answer: there is no difference between SEC and GPC, they are synonyms.
Moreover, of course you can do SEC/GPC system with a HPLC systems, MOST commercial SEC/GPC systems are nothing but modified commerical HPLC systems. The only really relevant difference are the columns and the detectors. For HPLC, UV-Vis detectors are THE standard, for GPC/SEC differential refractive index detector are THE standard. For GPC/SEC viscosimetry and/or light scattering makes sense, too, depending on your analyte also UV-Vis.
