Hello. I need to incubate HEK 293 T cells with a molecule that interfere with serum. So, is it possible to make this incubation in DMEM without serum during 4 days?
Yes, it is....we produce virus in SF medium and they do not particularly like it, but they survive....there's serum-free medium available e.g. OptiMEM that may be an healthier (although more expensive) alternative to the DMEM.
yeah i agree with Christoph. OMEM is recommended in cases of transfection as well, which ranges from 24-48 hours, even 72 hours in certain cases. I sugest you buy OMEM. Gibco OMEM is the best.
Yes, HEK293 cells can survive in DMEM without serum. We also use them to produce proteins and have optimized them to grow without serum. Initially they used to detach but on optimization we have pretty much got them used to grow in serum free Media. Also have a look at these alternatives, non-animal derived components - http://www.ncbi.nlm.nih.gov/pmc/articles/PMC3284993/
You can also grow them in Hyclone SFMTransfx, in Freestyle 293 and Excell medium. Following the article suggestion from Stanley Kimani, here is the complete paper in which you can see the performance of the HEK 293 cells in the media mentioned above and also after Freestyle 293 medium optimization.
