I am looking for a good marker of NFkB activation in order to analyze it in lymphocytes obtained from inflamed mice by flow cytometry. Would antibodies to phophorylated IkB alpha, such as
Phospho-IkB alpha (Ser32, Ser36) Monoclonal Antibody (RILYB3R), PE, eBioscience™, be suitable? And should I use simple permeabilization buffer (the one we use for cytokine staining) or buffer for nuclear transciption factors. I suppose i should use first one because the process of IkB phosphrylation takes place in the cytosol.
Dear, the best one would be NF-κB p65. Better when performed on homogeneous cells after purification.
