I am receiving a cancer cell line from a collaborator that is not commercially available from ATCC. Is there a way to verify the tissue of origin and the histological subtype? I know you can do STR analysis but only for cell lines that have known STR data which is not the case with this cell line. I am thinking of preparing a cell block and performing IHC for common markers characterizing this cancer type. I would appreciate any other ideas.
Thank you both. Is there a genetic method? I know that sometimes journals ask for cell verification by STR before publishing. What would be the policy on that if that information is not available?
Is it human or murine cell line? Is it still tumourogenic then it is quite simple. If it is a murine cell line then inject it into syngenic mice and check the tumour grown - You authenticate this way the genotype (it grows on syngenic mice) and tumour type (just make some histological slides of the tumour). With human tumour cell lines You can check the second aspect by injecting it into SCID mice. Of course IHC works also and with the tumour cell lines which do not induce tumour growth in vivo.

Historically, cell-line authentication testing had been performed using isoenzyme analysis and karyotyping.
Another test which can be used is single nucleotide polymorphisms (SNPs).

maybe You can ask them if STR authentication of Your cell line is possible (it could be unpublished by the firm).
I do have similar problems when we got new non-commercial cell lines or when we derived a new cell line from patients.
I do agree with previous suggestions. Following are suggestions based on our lab experience:
1) Nevertheless, we always perform STR profiling even though its reference not available. We do STR profiling beginning of the use of new cell lines and also after 6 month to 1 yr, to show the genomic integrity remains similar and Journals accept that.
2) Immunohistochemistry ( preferably florescence) for epithelial markers or other depends tissue of origin
3) In-vivo tumorigenecity test and compare it with other known cancer cell line or on-cancerous cell line of same origin.

Please to STR analysis first and try injecting to synergic mice, then you can verify totally especially in case of murine cell line (allograft) but with less successful for human cancer cell lines (Xenograft). TQ
