I am analyzing the concentration of nitric oxide (NO) in culture medium after exposing myofibroblasts of colon CCD-18Co to LPS and IL-1b. As the protocol of the kit recommends to use culture medium without phenol red, and the fetal bovine serum could influence the results, the treatment was made in non-phenol red medium (MEM) and at 0.1% of fetal bovine serum. However, when the medium was analyzed using the commercial kit from Sigma, there were no differences between control and treated.
Recently, it has been described that the exposition of these cells to LPS increases the production of NO (Zhu et al., 2012)

Is it possible that the treatment of the cells with LPS and IL-1b in culture medium at 0.1% of serum avoid seeing the effect of these compounds on the production of NO? Is it necessary to make the treatment in medium at 10%?
