I"m induced my rice plant numbers of chromosomes (2n=21) using chemical mutugenesis colchicine to double the number of chromosome of my paddy plant and now i want to use flow cytometry machine to confirm that my paddy plant already been mutated or not ?

An internal standard is a plant sample of a known and characterized species you add to the sample you want to measure.
An example I give you that can match yours : we want to measure the genome size variability in sunflower. We take our sunflower collection, and as as standard we will use the pea Pisum sativum "Express Long".
Pea nuclei will form a peak, sunflower ones will form another and by comparison between the two peaks, you can measure the genome size of the sunflower lines, or your rice lines, and deduce the mutated ones.
