A. baumannii is a lactose non-fermenter bacteria, I cultured clinical swab on hiCrome Acinetobacter agar Base, then subculture on MacConkey
because this bacteria is lactose non-fermenter, it should give a media with yellow color, but in sample the MacConkey still with pink color
is this normal??
What is the pigmentation of your bacterium like? Does it produce pink colour on general purpose medium, say tryptic soy agar? What is the colour of hiChrome medium? There are different kinds of hiChrome agar. Which did you use? If hiChrome medium was red to pink and the transfer was heavy, chances are that the A. baumannii got its pink colour from the previous medium This is one of the reasons for washing of cultures in microbiology. It could also be a mutant that may have picked up the gene for lactose-fermentation while growing in mixed culture. All these are speculations. Repeat your experiment several times by growing the bacterium in a general purpose medium say plate count agar or tryptic soy agar, after washing. Watch out for the pigmentation referred to. Something very interesting may well be going on. So, take your time and follow your research carefully. These things happen.

dose the hiCrome agar for A. baumannii destroy the bacterial gene ... so when we did the subculture on Macconkey, it dose not convert to yellow color and the media still pinkish, because the bacteria could be destroyed ??
