I will be transporting HEKa cells for irradiation 90 minutes away and back. I will be using CO2 independent media from life technologies in 50mL conical tubes. At what temperature should I transport the cells?
Also, the protocol states that the media should be supplemented with L-glutamine and FBS. Is this supplementation required for short periods or only for growing without a CO2 incubator?
Temperature and CO2 are different things. CO2 is to keep pH in bicarbonate-buffered media. So ideally at 37C, but check with technical support of the company from where you bought the cell line or who supplied to carbon dioxide independent medium.
1st answer: CO2-Independent Medium will maintain a pH of 7.3 to 7.5 over a 72-hour test period; there is no relation to temperature; so, you can keep your cells at normal growth temperature (37C).
2nd answer: CO2-Independent Medium requires supplementation with both serum and L-glutamine prior to use.
